PCR Master Mix Calculator is a web-based calculator for PCR dilution, reagent scaling, and master mix preparation. It uses the C1V1 = C2V2 dilution equation to calculate the stock volume required to reach a specified final concentration and reaction volume. Users can enter the target reaction volume, reaction count, and overage margin to calculate the total master mix required for multiple reactions. The calculator supports scaling for common plate formats, including 96-well and 384-well workflows. An auto-water balancing function calculates the required nuclease-free water after accounting for the volumes of other reagents. Users can exclude template DNA from the bulk master mix so it can be added separately to individual wells. Custom reagents can also be added for multiplex PCR setups involving additional primers or probes. The tool provides a preparation summary and bulk tube recipe that can be used while assembling the master mix.
